Title Of Talk: “Epigenetic Regulation of Cardiac and Vascular Diseases”

Summary Of Seminar:  Hepatic vascular cavernomas, the most common benign tumor of the liver, cause lethal complications such as hepatic rupture, consumption coagulopathy, and cardiac failure.  Although Virchow and Frerichs described hepatic vascular cavernomas as a distinct clinical entity
in the mid-1800s, their genetic etiology, molecular mechanism, and effective treatment remain undefined. Recent study from my laboratory identified gain-of-function mutations in KRAS or BRAF genes within liver endothelial cells as a causal mechanism for hepatic vascular cavernomas.  Recent work from my laboratory has established a critical interplay among chromatin modifying enzyme, transcription factor, and signaling pathways during pathogenesis of congenital heart disease.  Our recent study identifies a causal relationship between Congenital Heart Disease, the most common developmental defect in children, and defective developmental energy generation.  We demonstrated that two class I histone deacetylases, Hdac1 and Hdac2, silence cryptic
transcription to promote mitochondrial function in developing murine hearts.
